The present work aims to establish a protocol for in vitro propagation of Ocimum americanum using inflorescence, leaf, nodal and internodal explants. Embryogenic callus was induced from all explants on MS medium fortified with 1 to 3.5 mg/l of 2,4-D. Optimum frequency of somatic embryogenesis was found at 2.5 mg/l 2,4-D in inflorescence, leaf, nodal and internodal explants respectively. Regeneration and shoot elongation was obtained when transferred this calli into different concentrations of BAP and kinetin. Of various concentrations of BAP and kinetin used in the present study 2.5 mg/l BAP + 2 mg/l kinetin was found to be more effective for shoot regeneration and elongation. Rooting of shoots was achieved on half strength MS medium containing 2 mg/l NAA. The regenerated plantlets were acclimatized and successfully transferred to field conditions.
