Abstract

An efficient protocol was developed for inducing callus and plant regeneration using petiole explants. The calli were raised on MS medium supplemented with different concentrations and combinations of auxins and cytokinins. The highest percentage of green compact callus induction (89.6 %) as well as proliferation was observed on MS medium containing 2,4 - D (2.0 mg/l) and BAP (1.0 mg/l). The maximum percentage of shoot bud formation (82.2%) was obtained on MS medium fortified with the combination of BAP (3.0 mg/l) and IAA (1.0 mg/l). The regenerated shoots were cultured on rooting medium containing MS basal medium with various concentrations of IBA and the highest rooting percentage noticed was 85.0%. The rooted plants were transferred to plastic cups for hardening. The regenerated plantlets were morphologically uniform and exhibited normal seed set.
